Gene silencing spurs fountain of youth in mouse brain

Feb. 7, 2013 ? Cognitive decline in old age is linked to decreasing production of new neurons. Scientists from the German Cancer Research Center have discovered in mice that significantly more neurons are generated in the brains of older animals if a signaling molecule called Dickkopf-1 is turned off. In tests for spatial orientation and memory, mice in advanced adult age whose Dickkopf gene had been silenced reached an equal mental performance as young animals.

The hippocampus -- a structure of the brain whose shape resembles that of a seahorse -- is also called the "gateway" to memory. This is where information is stored and retrieved. Its performance relies on new neurons being continually formed in the hippocampus over the entire lifetime. "However, in old age, production of new neurons dramatically decreases. This is considered to be among the causes of declining memory and learning ability," Prof. Dr. Ana Martin-Villalba, a neuroscientist, explains.

Martin-Villalba, who heads a research department at the German Cancer Research Center (DKFZ), and her team are trying to find the molecular causes for this decrease in new neuron production (neurogenesis). Neural stem cells in the hippocampus are responsible for continuous supply of new neurons. Specific molecules in the immediate environment of these stem cells determine their fate: They may remain dormant, renew themselves, or differentiate into one of two types of specialized brain cells, astrocytes or neurons. One of these factors is the Wnt signaling molecule, which promotes the formation of young neurons. However, its molecular counterpart, called Dickkopf-1, can prevent this.

"We find considerably more Dickkopf-1 protein in the brains of older mice than in those of young animals. We therefore suspected this signaling molecule to be responsible for the fact that hardly any young neurons are generated any more in old age." The scientists tested their assumption in mice whose Dickkopf-1 gene is permanently silenced. Professor Christof Niehrs had developed these animals at DKFZ. The term "Dickkopf" (from German "dick" = thick, "Kopf" = head) also goes back to Niehrs, who had found in 1998 that this signaling molecule regulates head development during embryogenesis.

Martin-Villalba's team discovered that stem cells in the hippocampus of Dickkopf knockout mice renew themselves more often and generate significantly more young neurons. The difference was particularly obvious in two-year old mice: In the knockout mice of this age, the researchers counted 80 percent more young neurons than in control animals of the same age. Moreover, the newly formed cells in the adult Dickkopf-1 mutant mice matured into potent neurons with multiple branches. In contrast, neurons in control animals of the same age were found to be more rudimentary already.

Blocking Dickkopf improves spatial orientation and memory

Several years ago, Ana Martin-Villalba had shown that mice lose their spatial orientation when neurogenesis in the hippocampus is blocked. Now, is it possible that the young neurons in Dickkopf-deficient mice improve the animals' cognitive performance? The DKFZ researchers used standardized tests to study how the mice orient themselves in a maze. While in the control animals, the younger ones (3 months) performed much better in orienting themselves than the older ones (18 months), the Dickkopf-1-deficient mice showed no age-related decline in spatial orientation capabilities. Older Dickkopf-1 mutant mice also outperformed normal animals in tests determining spatial memory.

"Our result proves that Dickkopf-1 promotes age-related decline of specific cognitive abilities," says Ana Martin-Villalba. "Although we had expected silencing of Dickkopf-1 to improve spatial orientation and memory of adult mice, we were surprised and impressed that animals in advanced adult age actually reach the performance levels of young animals."

These results give rise to the question whether the function of Dickkopf-1 may be turned off using drugs. Antibodies blocking the Dickkopf protein are already being tested in clinical trials for treating a completely different condition. "It is fascinating to speculate that such a substance may also slow down age-related cognitive decline. But this is still a dream of the future, since we have only just started first experiments in mice to explore this question."

Marta Segal Block: What I Learned about Weddings from My Parents ...

My parents recently celebrated their 50th wedding anniversary. After much debate about the best way to honor it, they decided to host a party in a restaurant owned by a family friend. I was asked to give the toast. Despite the fact that I work with words and weddings and have given tons of advice on speeches and parties, I was flummoxed.

Normally, you would expect a toast at an anniversary party to focus on either the couple's relationship or the original wedding. But, my mother hates everything to do with her wedding. Believe me when I say that 50 years has just started to dull her pain at the problems with her wedding. So, the wedding itself was completely off-limits. My parents' relationship was also off limits for a speech. Clearly, over 50 years my parents have found something that works for them, but it's not exactly a story book relationship, unless that story book was written by Edward Albee. Let's just say, we're a family that has conversations at high volume, often with unprintable words, words certainly not appropriate for a public toast.

So, I started to think deeper about weddings and what they mean and why we have them and why it's so important not just to my parents, but to all of their friends and family, that they were celebrating their 50th anniversary. This is what I came up with and what I said. I think it applies beyond my own family.

"As many of you know, I make my living in the wedding business. It's sort of an ironic career because I spent over 30 years of my life avoiding marriage, and then spent the nine months that I was engaged asking my husband if we could elope. But, over the course of my years in the wedding business I've come to realize that weddings, not just marriage, are important.

Weddings are NOT, as the popular stereotype would tell us, just about the bride and groom. Except for elopements, weddings are a communal event and they are about community. I think that's why so many people care so much on both sides about marriage equality rights. We all want a say in how our communities are formed. For some of us, we want those communities formed on values of inclusion and love, others may want their communities formed according to their understanding of religious or historical traditions.

My parents care deeply about community. Not just because as professors and social scientists they study communities and how they are formed and changed, but because they care about their own community. They are actively and endlessly involved in their synagogue, in Girl Scouts, in professional groups, and in the wider community. Even the space we're standing in, Hillbilly Tea, is a testament to their love of community. Hillbilly Tea is owned by a dear friend of my sister's and mine from high school. My parents always welcomed all of our friends into their home. Anyone that we loved, my parents accepted as part of their community.

There are very few people here today who were at my parents' wedding 50 years ago. My aunt and uncles, my father's cousin, and his aunt. Those people represent the original blended community that happened when they were married. Their wedding combined their individual families and communities. Everyone standing here now is part of the community that my parents have worked so hard to build over the past 50 years.

This is why weddings and anniversaries are so important. They remind us that we are not just about two people, but about building a larger community. Tonight, we celebrate the community that my parents have built for the past 50 years. L'chaim!"

Did Selena Gomez Take Justin Bieber Back? (Please, No!)

Hmm, maybe Justin Bieber's heart-wrenching new breakup song is making Selena Gomez have second thoughts! On Saturday morning, Gomez, 20, was photographed leaving Bieber's house, escorted (in the nice way, not in the "get out of here" way) by Justin's bodyguard.

Alicia Keys performed a lounge-y, piano-tinged ? and live ? version of the national anthem ahead of the Super Bowl on Sunday.

The Grammy-winning singer played the piano as she sang "The Star Spangled Banner" in a long red dress with her eyes shut. Her publicist said the performance was live, days after halftime performer Beyonce admitted singing along to a prerecorded track at the Inauguration.

Keys' version was soft and featured additional lyrics: She added "living in the home" before belting "home of the brave" as she finished the song.

Before Keys hit the field, Jennifer Hudson performed "America the Beautiful" with the 26-member Sandy Hook Elementary School chorus, a performance that had some players on the sideline on the verge of tears.

The students wore green ribbons on their shirts in honor of the 20 first-graders and six adults who were killed in a Dec. 14 shooting rampage at the school in Newton, Conn.

The students began the song softly before Hudson, whose mother, brother and 7-year-old nephew were shot to death five years ago, jumped in with her gospel-flavored vocals. She stood still in black and white as the students moved to the left and right, singing background.

Keys and Hudson warmed up the field for Beyonce, who is set to perform at the half-time show.

Microsoft launches 4Afrika initiative with Huawei W1 variant, TV white space broadband project

Microsoft launches 4Afrika initiative with Huawei W1 variant, lowcost wireless broadband

Following the lead of co-founder Bill Gates Microsoft is taking more interest in Africa, announcing its 4Afrika Initiative with a stated aim of improving the continent's global competitiveness. There are several plans under way as a part of the project, with one of the first being a new Windows Phone 8 device from Microsoft and Huawei. Pictured above, the Huawei 4Afrika phone is a specially tailored version of the existing Ascend W1 meant as an affordable option (no price announced yet) for first time smartphone buyers that also comes preloaded with apps created by African developers for African consumers, and a subsection of the existing Windows Phone Store that will continue to focus on "locally-relevant" apps and content. It will be available in blue, red, black and white when it launches later this month in Angola, Egypt, Ivory Coast, Kenya, Morocco, Nigeria, and South Africa.

Another part of the push is a pilot project Microsoft is working on with the Kenyan government and Indigo Telecom which combines solar powered base stations using TV white space technology to offer affordable wireless internet access. Meant to bring broadband to places that currently lack even electricity, the deployment is called Mawingu, connecting a healthcare clinic and several schools in its initial test. After several years of pushing the tech, which takes advantage of unused TV broadcast spectrum, Microsoft hopes to convince other nations to make the legal/regulatory changes to start using it as well. There's a press release after the break with more details, as well as a video and more information available beyond the source links.
